My favorite is Boston. I've been to New York, philadelphia, pittsburgh, and they're nice. Boston though is an older city, has grown up around its history, and is very pedestrian friendly. From what I understand the economy there completely sucks, unless you're a multi-gazillionare, but it's still a neat city.
But it's very difficult to say which city I like best in the US. I tend to like the countryside around cities better than the cities themselves, like VA's wine country, for example.

Favorite city? Las Vegas, Nevada.
Of course, my experience and travels are quite limited, but given a choice, I'll pick Las Vegas.
